Carlos Brathwaite hit Ben Stokes for four consecutive sixes in the final over to seal the win for his side.
Marlon Samuels had carried the innings with an unbeaten 85, beating his own record for the highest score in a World T20 final before his partner took the reigns and obliterated the target of 19 from the final over.
England had earlier been reduced to 23-3 and 111-7 as Joe Root top scored with 54, backed with cameos from Jos Buttler (32) and David Willey (21) too take England to a final score of 155-9 after 20.
They join their female counterparts as World T20 champions and the under 19 team won their world cup earlier this year, making it a triple victory for the teams.
After being reduced to 11-3 in reply to England's 155-9, losing star man Chris Gayle, Samuels and, first with DJ Bravo - then Brathwaite, rebuild and kept his side in the game and with wickets in hand, allowing for a dramatic finish, seeing the West Indies defy expectation and lift the trophy again.
Despite the heartbreak for England, it was a much improved tournament for the young, fearless side, who saw their peers knocked out of the World cup last year in the group stages, leading to a complete overhaul of the one-day system by the ECB and Andrew Strauss.
The next World Cup is in England in 2019, with the next World T20 in Australia in 2020.
